Some SEGA game explore MODE button from 6-buttom gamepad:

Seems that mednafen 0.9.36.4 have no MODE button emulation.

Please add one!

Mednafen does support the Mode button. Change the input device to a 6-button gamepad (CTRL + SHIFT + 1 for the port 1), enter the button configuration mode (ALT + SHIFT + 1) and bind the Mode button to whatever key you want.
